ABSTRACT

@#Abstract: Objective To collect and organize malaria case data in Hubei Province from 2017 to 2021, compare and analyze the malaria epidemic characteristics on the before and after malaria elimination, and provide scientific support for Hubei Province to further optimize the comprehensive strategies to prevent re-transmission after the elimination of malaria. Methods The study was conducted by collecting the data of reported malaria cases of Hubei during 2017-2021 from the Infectious Disease Surveillance Reporting and Management System, and conducting the epidemiological characteristics of malaria on pre-elimination (2017-2019) and post-elimination (2020-2021). Results A total of 429 cases of imported malaria were reported in Hubei Province from 2017 to 2021, and the malaria epidemic showed an obvious trend of rising first and then falling. On the pre-malaria elimination, 374 malaria cases were reported, including 262 cases of P.falciparum (70.05%); on the post-malaria elimination, 55 malaria cases were reported, including 25 cases of P.falciparum (45.45%). There was a statistically significant difference in the proportion of infections caused by the four types of malaria parasites before and after the elimination of malaria (χ2=14.248, P<0.05). On the pre-malaria elimination, the peak of disease onset mainly occurred in January, July, and November; on the post-malaria elimination, the peak of disease onset mainly occurred in January to February, and December. Both before and after malaria elimination, the reported cases were mainly concentrated in Wuhan, Yichang, Huangshi, Xiangyang, Shiyan and Huanggang, but the range of cases showed a clear trend of narrowing. Before and after malaria elimination, malaria cases in Hubei Province were mainly among young and middle-aged males aged 30-49. The proportions of workers and migrant workers increased from 37.70% and 9.09% before the elimination to 50.91% and 18.18% after the elimination, respectively, with a statistically significant difference (χ2=17.839, P<0.05). The percentage of interval from onset of illness to initial diagnosis ≥ 5d decreased from 21.66% before the elimination to 10.91% after the elimination (χ2=6.448, P<0.05). The percentage of definitive diagnosis of malaria at initial diagnosis in town clinic increased from 18.18% before the elimination to 50.00% after the elimination. The proportion of malaria cases diagnosed by county-level medical institutions increased from 22.73% before the elimination to 34.55% after elimination. There was no statistically significant difference in the proportion of malaria cases diagnosed by medical institutions at all levels before and after the elimination of malaria (χ2=5.630, P>0.05). The proportion of cases with the interval between initial diagnosis and diagnosis within 24h increased from 43.85% before the elimination to 70.91% after the elimination. There was a statistically significant difference in the proportion of cases with the interval between initial diagnosis and diagnosis before and after the elimination of malaria (χ2=14.006, P<0.05). Before and after malaria elimination, all reported cases were mainly imported from African countries. Conclusions There are imported malaria cases reported every year in Hubei Province before and after the elimination of malaria, which poses a great challenge to the prevention of re-transmission. Therefore, it is necessary to strengthen the surveillance system, detect and standardize the treatment of imported malaria cases in a timely manner, conduct targeted retransmission risk surveys and assessments, and consolidate the achievements of malaria elimination.

@#Abstract: Objective To evaluate the completion and final effect of key parasitic disease prevention and control planning tasks in Hubei Province from 2016 to 2019, summarize the experience, find out the problems, and provide the basis for the next stage of prevention and control. Methods According to the requirements of the Final Evaluation Plan of the National Plan for the Prevention and Control of Hydatid Disease and Other Major Parasitic Diseases (2016-2020), a retrospective survey method was adopted to collect relevant data on the implementation and safeguard measures of the prevention and control of major parasitic diseases, and population infection status in Hubei Province in 2016-2019. Results From 2016 to 2019, We carried out 2 920 992 person times of publicity and education, 209 times of prevention and control technology training, 7 680 person times of business training, with an average of 52 sessions and 1 920 person times per year. We have allocated 3.445 2 million yuan for the prevention and control of parasitic diseases, including 1.722 2 million Yuan froom provincial government, to achieved full coverage of safe drinking water in rural areas under the current national standards, and 7.687 9 million harmless toilets have been built in rural areas. From 2016 to 2019, we carried out 39 658 person times of monitoring and disease investigation, the infection rate of human liver fluke was 0, and the infection rate of soil transmitted nematode was 0.42%. While the annual infection rates varied, there was no statistically significant difference in infection rate between years (χ2=2.276, P>0.05), but there were statistically significant differences in the infection rates between various soil nematodes (χ2=112.807, P<0.01). From 2016 to 2019, a total of 5 393 people were detected at 17 monitoring points, with the serum positive rate of 3.93% for paragonimiasis, there was a statistically significant difference in serological positive rate between years (χ2=146.011, P<0.01); a total of 738 stream crabs were collected, and the infection rate of intermediate host was 16.26%, wtih a statistically significant difference in the infection rate of stream crabs between years (χ2=49.731, P<0.01). Conclusions From 2016 to 2019, we adhered to the prevention and control strategy of "prevention first, prevention and control combined", implemented comprehensively various prevention and control measures, and achieved remarkable results in Hubei Province. The key parasitic diseases have been in a low epidemic situation, meeting the requirements of the prevention and control objectives. But the transmission risk still exists, the next step is to continue to strengthen security and monitoring and consolidate the achievements of prevention and control.

ABSTRACT

Objective@#To analyze the current nutritional composition of commonly consumed prepackaged foods among children in Chengdu and to provide a scientific basis for health education among children and adolescents.@*Methods@#Based on the 3 day and 24 hour dietary data of children aged 6-12 in Chengdu of the Southwest China Childhood Nutrition and Growth Cohort from 2021 to 2022, the nutritional information of prepackaged foods was collected by combining offline and online methods. All foods were classified step by step, and the nutrient content of each 100 g or 100 mL food was counted and graded.@*Results@#A total of 1 902 children s prepackaged foods in 23 sub categories of 10 major categories were investigated. Nuts and seeds, snack foods, instant foods and other dairy products had higher total energy(2 476, 2 027, 1 728, 1 816 kJ/100 g), with the nutrient reference value percentage(NRV%) exceeding 20%. Fish, poultry, meat, eggs and their products had the highest protein content(22.8 g/ 100 g ) with an NRV% of 38%, nuts and seeds had the highest fat content(47.5 g/100 g) with an NRV% of 79%, confectionery and jelly had the highest carbohydrate content(82.1 g/100 g) with an NRV% of 27%, and seasoning flour products had the highest sodium content with an NRV% up to 118%. Seasoning flour products and instant foods were mostly high sodium, high fat and high carbohydrate food. Baked and puffed foods were almost high fat and high carbohydrate. Fish, poultry, meat, eggs, beans and their products were rich in protein but mostly high in sodium. Beverages and cold drinks were low in other nutrients except carbohydrate.@*Conclusion@#The nutrient content of various prepackaged foods commonly consumed by children in Chengdu are quite different. Most of the foods consumed by children are high sodium, high fat and/or high carbohydrate. Nutrition education should be strengthened to help children choose healthy foods.

ABSTRACT

The repair of bone defects, especially for the large segment of bone defects, has always been an urgent problem in orthopedic clinic and attracted researchers' attention. Nowadays, the application of tissue engineering bone in the repair of bone defects has become the research hotspot. With the rapid development of tissue engineering, the novel and functional scaffold materials for bone repair have emerged. In this review, we have summarized the multi-functional roles of osteoclasts in bone remodeling. The development of matrix-based tissue engineering bone has laid a theoretical foundation for further investigation about the novel bone regeneration materials which could perform high bioactivity. From the point of view on preserving pre-osteoclasts and targeting mature osteoclasts, this review introduced the novel matrix-based tissue engineering bone based on osteoclasts in the field of bone tissue engineering, which provides a potential direction for the development of novel scaffold materials for the treatment of bone defects.

ABSTRACT

The abnormality of ubiquitin proteasome pathway is an important factor leading to the imbalance of protein homeostasis. In this process, the deubiquitinase responsible for removing the ubiquitin chain of protein substrate is very important. Its abnormal activity or expression can cause the functional changes of key oncogenic/tumor suppressor proteins, which directly or indirectly lead to the occurrence, development and malignant evolution of tumors. Based on this, the discovery and research of small molecule inhibitors targeting deubiquitinases have become a hot field of anti-tumor candidate drugs. This review will focus on the regulatory effect and mechanism of ubiquitin proteasome pathway, especially deubiquitinase on tumor, introduce the application of deubiquitinase small molecule inhibitors in tumor treatment, and discuss the research status and latest progress of small molecule inhibitors, so as to provide ideas for the research of new anti-tumor strategies based on deubiquitinase.

ABSTRACT

Objective@# To compare the stress distribution of different all-ceramic restoration materials and thicknesses in dental crown restorations using the finite element method and provide a reference for the selection and design of clinical crown restoration materials.@*Methods@#A finite element model of mandibular first molar implant crown restoration was created, and 6 crown thickness designs and 4 different crown restoration materials were evaluated, namely, resin-based ceramics (Lava Ultimate and Vita Enamic), lithium disilicate glass-ceramics (IPS e.max CAD), and zirconia ceramic (Cercon) designs. The mandibular first molars were loaded at 600 N, and the stress distribution was analyzed by using the finite element software ANSYS 10.0.@*Results@#The crown stress analysis showed that 156.05 MPa was the highest in 4 mm Cercon group and 18.85 MPa was the lowest in 1 mm Lava Ultimate group. The stress analysis of resin cement showed that 62.52 MPa was the highest in the 4 mm Lava Ultimate group and 16.74 MPa was the lowest in 1 mm IPS e.max CAD group. During the use of the finished platform, the stress concentration of the Lava Ultimate group in the crown prosthesis and resin cement was higher than that of the personalized platform with the same crown thickness.@*Conclusion@# With increasing crown thickness, the maximum principal stress concentration in crown restoration and resin cement increases. Personalized abutments are more conducive to reducing stress concentrations for resin-based ceramics.

ABSTRACT

Objective To evaluate the correlation of long non-coding RNA (lncRNA) antisense non-coding RNA in the INK4 locus (ANRIL) with disease risk and severity of bronchial asthma as well as plasma microRNA (miRNA)-125a expression in child patients. Methods Seventy children with asthma exacerbation, 70 children with asthma remission and 70 matched healthy controls were consecutively enrolled in this study. Laboratory parameters and lung function indexes of the participants were recorded. LncRNA ANRIL and miRNA-125a expression levels in plasma were determined using qRT-PCR. Tumor necrosis factor α (TNF-α), interleukin (IL)-1β, IL-6 and IL-17 levels in plasma were determined using enzyme-linked immunosorbent assay (ELISA). Results LncRNA ANRIL expression was the highest in the asthma exacerbation children, followed by asthma remission children and healthy controls. There were significant differences in the expression of lncRNA ANRIL among the three groups (all P<0.01). Receiver operating characteristic curves revealed that lncRNA ANRIL could well differentiate the participants in the three groups. In the children with asthma exacerbation, lncRNA ANRIL expression was associated with disease severity (P=0.001), positively associated with the levels of TNF-α, IL-1β, IL-6 and IL-17 (all P<0.05), while negatively associated with forced expiratory volume in one second (FEV1)/forced vital capacity (FVC) and FEV1 as percentage of predicted (FEV1%Pred) value (both P<0.05). LncRNA ANRIL expression was also positively associated with the levels of TNF-α, IL-1β and IL-17 in the asthma remission children and IL-6 level in healthy controls (all P<0.05). LncRNA ANRIL expression was negatively associated with miRNA-125a expression in all the participants (all P<0.05). Besides, miRNA-125a expression was positively correlated with FEV1/FVC and FEV1%Pred value in the children with asthma exacerbetion (both P<0.001). Conclusion LncRNA ANRIL may serve as a novel biomarker for predicting asthma, asthma acute exacerbation and severity, and inflammation level. It may participate in the development and progression of asthma in children via targeting miRNA-125a..

ABSTRACT

Objective:To investigate the effects of combination of transcranial direct current stimulation (tDCS) and mirror therapy (MT) on upper limb motor function after stroke. Methods:From April, 2018 to March, 2019, 78 stroke inpatients were randomly assigned into tDCS group (n = 26), MT group (n = 26) and combined group (n = 26). All the patients received their related therapy, respectively, based on routine medicine and rehabilitation. Their motor-evoked potential cortical latency (CL) and central motor conduction time (CMCT) in affected brain area were measured before and four weeks after treatment, while they were assessed with Fugl-Meyer Assessment-Upper Extremity (FMA-UE) and modified Barthel Index (MBI). Results:All CL, CMCT, and the scores of FMA-UE and MBI improved significantly in all the groups after treatment (|t| > 2.609,P < 0.05), and improved more in the combined group than in both tDCS group and MT group (P < 0.05). Conclusion:Both tDCS and MT could improve excitability of cerebral cortex of the affected brain area and promote the recovery of upper limb motor function for patient after stroke, and it is more effective in combination mode.

ABSTRACT

At present, most of Chinese patent medicines have problems, such as weak scientific basis, unclear clinical value evaluation, etc., which lead to the lack of sufficient evidence for the entry into the essential medicine list, medical insurance catalog and other important health resource allocation tools. This has a negative impact on the development of traditional Chinese medicine. It is urgent to establish a comprehensive evaluation methodology system of Chinese patent medicine in China. There are at least three reasons including producing evidence for clinical rational use scientifically, developing an objective and transparent selecting strategy, eventuating and highlighting its clinical value. The process and method of health technology assessment(HTA) can provide reference for the establishment of comprehensive evaluation method of Chinese patent medicine. However, HTA is rarely carried out in the field of traditional Chinese medicine. The latest domestic and international research showed that HTA combined with multi-criteria decision analysis(MCDA) can increase the transparency of decision-making and improve the quality of decision-making. The combined two formed the EVIDEM framework, which is conducive to drug selection and its clinical use. In this paper, the EVIDEM framework was introduced to establish the methodology framework of the comprehensive clinical evaluation system of Chinese patent medicine. A preliminary research idea has been put forward. In the future, we can establish a set of comprehensive clinical evaluation methods for Chinese patent medicine in cooperation with relevant national drug decision-making departments and management departments. It is believed that such cooperation could promote the full implementation of the re-evaluation for Chinese patent medicine.

ABSTRACT

Objective:Aphanamixis grandifolia,a perennial herb of genus Aphanamixis (Meliaceae),has effects in soothing activating collaterals,dredging paisy,expelling wind-evil and removing wetness. This paper aimed to investigate terpenoids from the stems and leaves of A. grandifolia and reveal the effective substances. Method:Totally 22 kg leaves and twigs of A. grandifolia were extracted with 90% EtOH for three times by heating reflux. These extracts were decompressed and concentrated,and then dissolved in water. The solvent was successively extracted with petroleum ether and ethyl acetate. The chemical constituents from petroleum ether and ethyl acetate fractions were isolated by macroporous,silica gel,Sephadex LH-20 and ODS columns,and their chemical structures were determined through MS,NMR analysis(1H and 13C-NMR)and spectroscopic data from literatures,respectively. Result:Twelve compounds were obtained and identified as 16α-hydroxy-3-oxo-24-methyllanosta-7,9(11),24(31)-triene-21-oic acid (1),23(E)-cycloart-en-25-ethoxy-3-ol(2),23(Z)-9,19-cycloart-ene-3β,25-diol(3),23(E)-cycloart-en-3β,25-diol(4),labda-8,13-(E)-dien-15-ol(5),labda-7,13-(E)-dien-15-ol(6),vulgarol(7),(S,E)-6-[6-(5,5-dimethyl-4-oxo-4,5-dihydrofuran-3-yl]-2-methylhepta-1,6-dien-1-yl]-4-methyl-5,6-dihydro-2H-pyran-one(8),nemoralisin(9),nemoralisin C(10),1S,4R,5S,6R,7S,10S-1(5),6(7)-diepoxy-4-guaiol(11) and 1S,4S,5S,10R-4,10-guaianediol(12),respectively. Conclusion:The structures involves triterpenoids,diterpenoids and sesquiterpennoids,and eight of them (1-3,5-8 and 12) are obtained from A. grandifolia for the first time. Those compounds are also isolated from the geneus Aphanamixis for the first time.

Three new sesquiterpene glycosides with alloaromadendrane and ylangene-derived type aglycones, named dendrofindlayanosides A-C (1-3), one new cyclopacamphane type sesquiterpene named dendrofindlayanobilin A (4), together with five known compounds have been isolated from stems of Dendrobium findlayanum. Their structures were determined on the basis of spectroscopic and chemical methods.

ABSTRACT

Objective:To investigate the effects of sling exercise therapy combined with Kinesio Taping on the balance and walking for stroke patients. Methods:From October, 2017 to August, 2018, 66 patients after stroke were randomly divided into routine group (n = 22), sling group (n = 22) and combined group (n = 22). All the groups accepted routine rehabilitation, while the sling group accepted sling exercise therapy based on routine group and the combined group accepted sling exercise therapy and Kinesio Taping. They were assessed with Berg Balance Scale (BBS) and Fugl-Meyer Assessment-Lower Extremities (FMA-LE), and their gait parameters were measured with Motion Analysis, before and four weeks after treatment. Results:There were obvious improvements in the BBS score, FMA-LE score and gait parameters in all the groups after treatment (P < 0.05), and it was ranked from better to worse as the combined group, the sling group and the routine group (P < 0.05). Conclusion:Sling exercise therapy combined with Kinesio Taping may further improve balance and walking for stroke patients.

The incidence of thyroid cancer has increased all over the world in recent years,which has become a hot issue of widespread concern.Surgery is an important means for the treatment of thyroid cancer.Standard surgical treatment can effectively improve the prognosis and quality of life of patients,while non-standard surgical treatment can increase the risk of recurrence,reduce the survival rate,and bring serious physiological,psychological and economic burden to patients.In order to standardize the diagnosis and treatment of thyroid cancer and improve the prognosis of patients,many guidelines have been issued at home and abroad.With the application of new therapies and the emergence of new clinical evidences,guidelines are constantly updated in various countries,the most representative of which are the guidelines issued by the American Thyroid Association and the National Comprehensive Cancer Network.In the past,surgeons paid more attention to the European and American guidelines,but little was known about the treatment of thyroid cancer in neighbouring Japan. Following the publication of the first edition of Japanese Clinical Practice Guidelines for Thyroid Tumors in 2010,Japan formally released a revised edition of the old edition of the guidelines in 2018. Compared with the old version,the recommendations of the new version are more comprehensive and accurate,including risk stratification of papillary cancer and indications of primary lesion resection and lymph node dissection in patients with different risk stratification,principles of primary and complementary surgery for follicular carcinoma of different histological types,and principles of treatment for hereditary and sporadic medullary carcinoma,etc.

ABSTRACT

OBJECTIVE: To explore the clinical characteristics,and discuss the prevention of lateral neck lymph node reoperation.METHODS: The clinical data of 31 cases of lateral neck lymph node reoperation performed between January2013 and December 2017 in the First Affiliated Hospital of China Medical University were analyed retrospectively.RESULTS: All 31 patients accepted 1 to 3 times lateral neck lymph node dissection,but the extent of lymph node dissection was not described enough clear in 17 cases. Twenty-four of 31 cases were found within 12 months from last operation to abnormal lymph nodes. In imaging diagnosis,the rate of metastasis in level Ⅳ was 58.1%,level Ⅱ(outside the outer edge of internal jugular vein)was 41.9%,trigonum caroticum and level Ⅲ(outside the outer edge of internal jugular vein)were 22.6%,lymph node between sternocleidomastoid and sternohyoid muscle(LNSS)was 12.9%. In pathology after surgery the rate of metastasis at levels Ⅱ,Ⅲ,Ⅳ,Ⅴb,LNSS was 64.0%,81.8%,68.4%,8.3%,13.6% respectively.CONCLUSION: The imaging assessment has significant effects on lateral neck lymph node dissection,within standardized procedure in surgery,which should help reduce the reoperation caused by human factor.

ABSTRACT

Objective To observe the effect of breathing training on postpartum stress urinary incontinence (SUI) based on pelvic floor muscle exercise and biofeedback electrical stimulation. Methods From July to October, 2017, 64 SUI patients at the sixth week after childbirth were randomly divided into con-trol group (n=32) and observation group (n=32). The control group was treated with pelvic floor muscle exercise and biofeedback electrical stimulation. The observation group was supplemented with breathing training addition-ally. The pelvic floor muscle myoelectric value, urinary incontinence frequency, one hour urine pad test and the average thickness of the transversus abdominis were evaluated before and twelve weeks after treatment. Results Before treatment, there was no significant difference in the pelvic floor muscle myoelectric value, urinary incon-tinence frequency, one hour urine pad test and the average thickness of the transversus abdominis between two groups (P>0.05). Twelve weeks after treatment, the pelvic floor muscle myoelectric value significantly increased in both groups (t>12.564, P<0.001), and was higher in the observation group than in the control group (t=14.064, P<0.001); the urinary incontinence frequency and the positive rate of one hour urine pad test significantly de-creased in both groups (Z>9.615, P<0.001), and were lower in the observation group than in the control group (Z=-2.767, χ2=6.564, P<0.05); the average thickness of the transversus abdominis had no improvement in the control group (t=-1.510, P>0.05), and significantly increased in the observation group (t=-10.681, P<0.01), which was higher in the observation group than in the control group (t=7.541, P<0.001). Conclusion On the basis of pelvic floor muscle exercise and biofeedback electrical stimulation, supplementation with breathing training could reduce the symptoms of postpartum stress urinary incontinence, and improve the func-tion and the quality of life.

ABSTRACT

Combined hepatocellular-cholangiocarcinoma (CHC) is a mixed tumor containing elements of both hepatocellular carcinoma (HCC) and cholangiocarcinoma (CC).Its remarkable histological heterogeneity has been linked to putative hepatic progenitor cell (HPC) origin.However,detailed histological or phenotypic description is rarely documented.In the present study,we reassessed 68 cases previously diagnosed as hepatitis B-related CHCs by immunohistochemistry and double-fluorescence immunostaining,focusing on HPC associated phenotypic observation of intermediate area of the tumor.It was found that tumor cells showed remarkable heterogeneity in intermediate area.Tumor cells with intermediate morphology between hepatocytes and cholangiocytes were oval-shaped and small with scant cytoplasm and hyperchromatic nuclei,arranging in solid nests mostly.By Keratin 7 (K7) staining,it appeared that the nests of tumor cells represented a maturation process from the undifferentiated small cells to mature hepatocytes through the "transitional" cells.Then,these small cells were further confirmed with intermediate phenotype as HPC by exploring immature hepatocellular marker and HPC/biliary markers co-localization.In conclusion,the HPC associated trait in CHC can be interpreted by HPC origin or gain of"stemness" by dedifferentiation.It is still too soon to give a final word that it is innate or acquired signature of HPC associated trait in CHC.

Objective@#To evaluate the effect of palatal free gingiva in anterior maxillary defect restoration after epulis resection. @*Methods@# 22 cases were included in this study. Palatal free gingival flap was prepared to restore anterior maxillary defect after epulis resection. Clinical effect was evaluated according to gingival margin, gingival papilla index and modified sulcus bleeding index. @*Results @#Satisfactory clinical effect was achieved in all 22 cases, with adequate height, thickness, fullness and texture. @*Conclusin @#Palatal free gingival flap was clinically effective in anterior maxillary defect treatment after epulis resection, with satisfactory aesthetic clinical effect.

ABSTRACT

Objective @#To investigate the relationship between the level of the soluble triggering receptor expressed on myeloid cell (sTREM-1) in the gingival crevicular fluid (GCF) of individuals and peri-implantitis.@*Methods@#46 patients (75 implants) with different severities of peri-implantitis and 18 patients (75 implants) without peri-implantitis were selected in this study. The concentrations of sTREM-1 in GCF with different types of peri-implantitis were detected by enzyme-linked immunosorbent assay (ELISA). The peri-implantitis peroxidase depth (PPD), the distance from the shoulder of the implant to the bottom of the bony defect (DSB), the modified sulcus bleeding index (mSBI) and the modified plaque index (mPLI) were recorded. The correlation between PPD, DSB, mSBI, mPLI and sTREM-1 was analyzed. @*Results @#The concentrations of sTREM-1 in GCF in mild, moderate and severe peri-implantitis group were significantly higher than those in healthy group (P < 0.05). The concentrations of sTREM-1 in GCF in severe peri-implantitis group were significantly higher than those in mild and moderate group (P < 0.05). There was no significant difference in the concentration of sTREM-1 in GCF between mild peri-implantitis group and moderate group (P > 0.05). In addition, there was a significant positive correlation between the concentration of sTREM-1 and PPD, DSB, mSBI, mPLI.@*Conclusions @#The concentration of sTREM-1 in GCF is closely related to the severity of tissue inflammation around implant.

ABSTRACT

OBJECTIVE To investigate the develop mental toxicity of muscone to embryos. METHODS With zebrafish embryos as a model,The 3 h post fertilization (hpf)embryos were exposed to muscone at 5,10,20,40,80 and 160 μmol·L -1 culture solutions for 96 h and inspected daily with mi-croscopy for larval morphology.The drug solution was replaced every 24 h.Spontaneous move ments were checked at 24 hpf.Heart rate at 48 hpf,hatching rate,e mbryo deformity rate and mortality rate were evaluated.The expression of sepn1 was determined with real-ti me quantitative PCR technique at 96 hpf.RESULTS The 24 hpf spontaneous move ments showed no significant difference.At 48 hpf, spine curvature,pericardial ede ma,yolk sac ede ma,and abnormal swi mming were observed.In addition, the 48 hpf heart beats(10 s)was decreased fro m 26.5 ±1 .0 to 18.0 ±1 .9(P <0.01 ).At 48 hpf , hatching rate of 5 ~40 μmol·L -1 decreased(P <0.05),while of 160 μmol·L -1 increased (P <0.05) co mpared with muscone 0 μmol·L -1 .Muscone had little effect on hatching rate at other ti me points;Mal-formation rate and mortality rate at higher concentrations were up to 100%.The sepn1 gene expression at 96 hpf in the exposure groups decreased co mpared with that of control group(P <0.01 ).CONCLU-SION Muscone had toxic effects on the develop ment of zebrafish embryos,including spine curvature, abnormal swi mming,and pericardial ede ma.These effects may be related to the inhibition of sepn1 gene expression by muscone.
